On the network it is Stratford (Central, Jubilee, Elizabeth and the DLR) to Charlton (National Rail), plus a 6-minute walk at the far end. The catch is that it is a limited service, and weekend engineering works are common. A car does the whole thing door to door in about 20 min, or 30 min when the roads are full - and it does it with your bags in the boot. If you are travelling light and early, the train usually wins on price. If you are in a group, carrying anything, or leaving after an event, the car wins on everything else.
From the E6, E7, E13, E15, E16 streets the route runs via the A13 then the A12, finishing on A102 then A206 Woolwich Road. Roughly 5.1 miles, and the timing swings with the hour rather than the route: 20 min clear, 30 min loaded.

If you were thinking of driving yourself: The Valley sits inside a matchday or event-day controlled parking zone, on-site spaces are pre-book only and go early, and the residential streets around it are permit-holders only with active enforcement. Pre-booked both ways, none of that is your problem.
Doors are usually an hour before the match. Working back, that means leaving Newham around 30 min plus that hour on a busy night - and booking the return at the same time, because the road out is slower than the road in.
