By rail you would go Vauxhall (the Victoria line) across to North Greenwich (the Jubilee line), then a 5-minute walk at the far end. The catch is that the Jubilee platforms are a crush for 40 minutes after a show empties. Door to door by road is about 25 min, or 45 min when the roads are full, with no changes, no stairs and no queue at the barrier. It is a fair fight on the way there. On the way back, when several thousand people want the same platform, it stops being one.
From the SW8 streets the route runs via the A3036 then the A202, finishing on A102 Blackwall Tunnel Approach, then Millennium Way. Roughly 8.2 miles, and the timing swings with the hour rather than the route: 25 min clear, 45 min loaded.
Vauxhall pickups take in all of SW8. Controlled parking zones and one-way systems are the driver's problem to solve, not yours.

If you were thinking of driving yourself: The O2 Arena sits inside a matchday or event-day controlled parking zone, on-site spaces are pre-book only and go early, and the residential streets around it are permit-holders only with active enforcement. Pre-booked both ways, none of that is your problem.
Doors are usually an hour before the gig. Working back, that means leaving Vauxhall around 45 min plus that hour on a busy night - and booking the return at the same time, because the road out is slower than the road in.
