On the network it is Shoreditch High Street (the Overground) to North Greenwich (the Jubilee line), plus a 5-minute walk at the far end. The catch is that the Jubilee platforms are a crush for 40 minutes after a show empties. The taxi is about 20 min, or 40 min when the roads are full, but it is one journey rather than three, and nobody stands up for any of it. If you are travelling light and early, the train usually wins on price. If you are in a group, carrying anything, or leaving after an event, the car wins on everything else.
From the EC2A, E1, N1 streets the route runs via the A10 then the A1202, finishing on A102 Blackwall Tunnel Approach, then Millennium Way. Roughly 6.6 miles, and the timing swings with the hour rather than the route: 20 min clear, 40 min loaded.

If you were thinking of driving yourself: The O2 Arena sits inside a matchday or event-day controlled parking zone, on-site spaces are pre-book only and go early, and the residential streets around it are permit-holders only with active enforcement. Being dropped at the door and collected at an agreed point removes the whole problem.
Doors are usually an hour before the gig. Working back, that means leaving Shoreditch around 40 min plus that hour on a busy night - and booking the return at the same time, because the road out is slower than the road in.
