By rail you would go Whitechapel (District, Elizabeth and the Overground) across to North Greenwich (the Jubilee line), then a 5-minute walk at the far end. The catch is that the Jubilee platforms are a crush for 40 minutes after a show empties. The taxi is about 20 min, or 35 min when the roads are full, but it is one journey rather than three, and nobody stands up for any of it. Two people with hand luggage should take the train. Three or more, or anyone with a case, and the fixed fare splits better than the tickets do.
Out of Whitechapel (E1) the driver picks up the A11 then the A1202, joining A102 Blackwall Tunnel Approach, then Millennium Way for the run-in. That is about 5.3 miles. Reckon 20 min off-peak and 35 min if you are crossing town at the wrong moment.
Whitechapel pickups take in all of E1. Controlled parking zones and one-way systems are the driver's problem to solve, not yours.
Private hire pickup is next to Car Park 3, past the Tesco on Millennium Way - not the main Entrance A forecourt, which is coach and black-cab only. The Blackwall and Silvertown tunnels are free between 22:00 and 06:00, so a return leg after a late finish costs the same as the outbound. Your driver sends the exact standing point and the registration before they set off, so you are not circling a car park looking for a number plate.

If you were thinking of driving yourself: The O2 Arena sits inside a matchday or event-day controlled parking zone, on-site spaces are pre-book only and go early, and the residential streets around it are permit-holders only with active enforcement. Pre-booked both ways, none of that is your problem.
Doors are usually an hour before the gig. Working back, that means leaving Whitechapel around 35 min plus that hour on a busy night - and booking the return at the same time, because the road out is slower than the road in.
