By rail you would go Hackney Central (the Overground) across to North Greenwich (the Jubilee line), then a 5-minute walk at the far end. The catch is that the Jubilee platforms are a crush for 40 minutes after a show empties. The taxi is about 20 min, or 40 min when the roads are full, but it is one journey rather than three, and nobody stands up for any of it. It is a fair fight on the way there. On the way back, when several thousand people want the same platform, it stops being one.
The usual line from Hackney is the A107 then the A10, then A102 Blackwall Tunnel Approach, then Millennium Way at the end. About 6.5 miles in all - 20 min when it flows, 40 min when it does not.

If you were thinking of driving yourself: The O2 Arena sits inside a matchday or event-day controlled parking zone, on-site spaces are pre-book only and go early, and the residential streets around it are permit-holders only with active enforcement. A drop-off and a booked return sidesteps it entirely.
Doors are usually an hour before the gig. Working back, that means leaving Hackney around 40 min plus that hour on a busy night - and booking the return at the same time, because the road out is slower than the road in.
