By rail you would go Mile End (Central, District and Hammersmith & City) across to Wembley Park (Metropolitan and Jubilee), then a 12-minute walk at the far end. The catch is that Olympic Way is queue-managed after full time and the walk is uphill. Door to door by road is about 35 min, or 1h 05m when the roads are full, with no changes, no stairs and no queue at the barrier. It is a fair fight on the way there. On the way back, when several thousand people want the same platform, it stops being one.
The usual line from Tower Hamlets is the A13 then the A1261, then A406 North Circular, then Harrow Road and South Way at the end. Roughly 15.5 miles, and the timing swings with the hour rather than the route: 35 min clear, 1h 05m loaded.

If you were thinking of driving yourself: Wembley Stadium sits inside a matchday or event-day controlled parking zone, on-site spaces are pre-book only and go early, and the residential streets around it are permit-holders only with active enforcement. Pre-booked both ways, none of that is your problem.
