By rail you would go Mile End (Central, District and Hammersmith & City) across to Custom House (Elizabeth line and the DLR), then a 5-minute walk at the far end. The catch is that it is fine with one bag and awkward if you are carrying exhibition kit. Door to door by road is about 15 min, or 30 min when the roads are full, with no changes, no stairs and no queue at the barrier. Two people with hand luggage should take the train. Three or more, or anyone with a case, and the fixed fare splits better than the tickets do.
From the E1, E2, E3, E14 streets the route runs via the A13 then the A1261, finishing on A13 then Connaught Bridge and Western Gateway. That is about 4.7 miles. Reckon 15 min off-peak and 30 min if you are crossing town at the wrong moment.
Tower Hamlets pickups take in all of E1, E2, E3, E14. Controlled parking zones and one-way systems are the driver's problem to solve, not yours.
