Per vehicle, not per person. Tolls, congestion and crossing charges are already inside the price, and it does not change on the day.
By rail you would go Bank (Central, Northern, Waterloo & City and the DLR) across to Custom House (Elizabeth line and the DLR), then a 5-minute walk at the far end. The catch is that it is fine with one bag and awkward if you are carrying exhibition kit. Door to door by road is about 25 min, or 40 min when the roads are full, with no changes, no stairs and no queue at the barrier. Two people with hand luggage should take the train. Three or more, or anyone with a case, and the fixed fare splits better than the tickets do.
From the EC2, EC3, EC4 streets the route runs via the A10 then the A1211, finishing on A13 then Connaught Bridge and Western Gateway. That is about 7.5 miles. Reckon 25 min off-peak and 40 min if you are crossing town at the wrong moment.

Parking at ExCeL London is limited and the surrounding streets are metered or permit-only, so self-driving usually costs more in time than the fare saves. Being dropped at the door and collected at an agreed point removes the whole problem.
Doors are usually an hour before the event. Working back, that means leaving Bank around 40 min plus that hour on a busy night - and booking the return at the same time, because the road out is slower than the road in.
