The public-transport version starts at Mile End (Central, District and Hammersmith & City) and finishes at North Greenwich (the Jubilee line), with a 5-minute walk at the far end. The catch is that the Jubilee platforms are a crush for 40 minutes after a show empties. Door to door by road is about 15 min, or 25 min when the roads are full, with no changes, no stairs and no queue at the barrier. If you are travelling light and early, the train usually wins on price. If you are in a group, carrying anything, or leaving after an event, the car wins on everything else.
The usual line from Tower Hamlets is the A13 then the A1261, then A102 Blackwall Tunnel Approach, then Millennium Way at the end. About 3.0 miles in all - 15 min when it flows, 25 min when it does not.
Any address across Tower Hamlets works, E1, E2, E3, E14 included, at whatever hour the East London traffic is doing its worst.

The Blackwall and Silvertown tunnels are free between 22:00 and 06:00, so a return leg after a late finish costs the same as the outbound. Your driver sends the exact standing point and the registration before they set off, so you are not circling a car park looking for a number plate.

If you were thinking of driving yourself: The O2 Arena sits inside a matchday or event-day controlled parking zone, on-site spaces are pre-book only and go early, and the residential streets around it are permit-holders only with active enforcement. Being dropped at the door and collected at an agreed point removes the whole problem.
Doors are usually an hour before the gig. Working back, that means leaving Tower Hamlets around 25 min plus that hour on a busy night - and booking the return at the same time, because the road out is slower than the road in.
