On the network it is Wembley Park (the Metropolitan and Jubilee lines) to Custom House (Elizabeth line and the DLR), plus a 5-minute walk at the far end. The catch is that it is fine with one bag and awkward if you are carrying exhibition kit. The taxi is about 45 min, or 1h 15m when the roads are full, but it is one journey rather than three, and nobody stands up for any of it. It is a fair fight on the way there. On the way back, when several thousand people want the same platform, it stops being one.
Out of Wembley (HA0, HA9) the driver picks up the A406 then the A404 then the A4088, joining A13 then Connaught Bridge and Western Gateway for the run-in. Roughly 19.8 miles, and the timing swings with the hour rather than the route: 45 min clear, 1h 15m loaded.

Parking at ExCeL London is limited and the surrounding streets are metered or permit-only, so self-driving usually costs more in time than the fare saves. Pre-booked both ways, none of that is your problem.
Doors are usually an hour before the event. Working back, that means leaving Wembley around 1h 15m plus that hour on a busy night - and booking the return at the same time, because the road out is slower than the road in.
